Can I use this spot for engine temp sensor location?

I am not specifically sure yet which intake manifold this is but it is a 360 (not sure the specifics as of yet either). This car does have factory air so not easy to get around alternator where I guess the original sensor is located.

Wondering if I can use this spot for a aftermarket engine temp sensor.

I would say no. It looks like an auxiliary vacuum port location. Your temp sensor needs to go into a water jacket. That appears to be in the intake runner.

Yeah you are most likely right... I had a aftermarket intake on my last A body that had a 360 and my Duster is Gen3 Hemi swapped. So I am not that familiar with the original intake manifolds.
 
Usually the temp sender location is up close to the thermostat housing.
 
That is a vacuum port, and it is typically the spot for the vacuum tree that connects the brake booster and vacuum for the heater box doors.

Small block temperature sensor is located behind the AC compressor and typically next to the heater connection and OSAC bypass valve on the manifold.

See purple wire with 'L" shaped connector, that is the temperature sensor just behind the AC compressor bracket in this photo and next to the heater hose and OSAC bypass valve.
