I have a 1966 charger with a 383 no a/c, the alternator on it has a two groove pulley, does it matter if I replace it with a alternator that only has a single groove?

If it's a 383 with no A/C and has a double groove pulley, someone installed the wrong one. It should only have a single groove pulley, so to answer your question, no it won't matter as that's what it originally had.
