The hole is indeed related to a hot idle compensator valve system mounted to the back of the center carb if the list number called for it. Otherwise, the needed machine work for the passages, valve and cover would be omitted.

To answer the original question, yes, the '70 4144 with the enclosed idle bowl vent valve will work fine on the ’71 three nipple breather ECS system. No major functional difference between the ’70 California only N95 and the ’71 all federal N95 ECS systems.

I can guess, there is quite a bit of information presented in Uncle Tony’s video to disagree with IMO. Using a ECS fuel tank vent line as a return line? There is a reason the factory ran a separate return line on factory installed fuel pump vapor separator installations.
Not correct, perhaps you should have a looked at the factory service manuals from ’70-71, relating to the Evaporation Control System, it's function and the correct information on the fuel bowl vent valve hose routing.

The OP specified a manual transmission, had it been coded for N96, R-6193A would be the correct original list number. List number R-4735A is N96 automatic transmission only.
‘71 383 HP 4-speed, heated air(not seeing N96 on the tag), would have left the factory with a Holley 4160 List number R-6191A. Holley R-4668A is the list number for heated air/auto trans.
