How do I get this piece on? This is not even close to going on! Do I need to trim the drip rail? quarters and drip rail have been replaced

So because I have NO experience and I have replaced or removed EVERYTHING, I'm thinking I should put the door glass and quarter glass in and test the fitment, just to put my mind at ease. Might as well install the belt molding while I'm at it. I will also have to have the latches and strikers...

When doing final door gaps should the latches and strikers be on? should the weather stripping be installed? Does it make a difference? should I have the door glass in? What about when I am doing final bodywork? (filler) I assume you remove the glass, if so because of the weight of the glass...

Could it originally been seam sealer from the factory?
