I guess I'm having a senior moment. I worked for maw Mopar in the 60's and 70's and adjusted a ton of cable shifted 727's. But never a reverse manual valve body cable shifted trans. We always made adjustments while holding the column shifter in 1st gear. But with a reverse manual set up do I need to have the shifter in the D range or in 1st when making adjustments? The column shift indicator I have is PRN12D.

1965 Belvedere, TF 727 w/reverse manual valve body. Column shifter w/cables.
Service manual requires shifter be in 1st gear while adjustments are being made. This procedure is with a stock PRND21 indicator column or console shifter.
The column shifter indicator I have is PRN123. To adjust the cable, does the shifter need to be in the #1 position or the #3 position ?

I don't read directions and do mine this way. I leave the cable end off the trans shifter arm and put the shifter and trans in Park and check the alignment, then I put both (shifter and trans) in 1st and check it again if both check out attach the cable and call it done.

62 to 64 the service manual suggests holding in reverse. 65 suggests 1st gear. Either way you at the end of the shifter cable travel. In your case "D" would be correct.

After thinking about it for a couple days, I believe DVW is right.
Thanks
