I have a 522 stroker that has six pack with port Injection so 2 rails how is the return line to the tank run? Do I run two lines For the inlet and the Return line goes on the bottom of the fuel regulator?

pressure line goes in from one side of fuel rail ,then at the end of that rail. pipe to the other side and then return fuel from the regulator goes back to the fuel tank

So I don’t need a regulator that has a return I just need a regulator that regulates the fuel pressure?

I'm not sure but I think you still need a return after the regulator to dump the excess back to the tank. Many newer cars these days have the regular in the tank with the fuel pump. That way the manufacturers save $ not needing a return line from the engine AND the repair facilities make more money because they usually need to drop the tank to get at either. To make matters worse, the miserable S.O.B.s also put the fuel filter in the tank as well so they can soak more money out of everyone. They Suck!!!

Go to the Holley website and look at their EFI regulators. Read the instructions, they show how to install the regulator with fuel rails. You can probably use the 12-880 regulator but it depends on how much fuel flow you are expecting. You might want the next size up on that engine. Holley has a bunch of regulators so you shouldn't have any problem finding one that matches your needs.

On my race engine I tied the rails together at both ends and fed fuel in one end and took it out the other to the regulator. You can also loop it from one side to the other. The loop method is used on lower performance engines, tying both sides together is used on the higher hp setups.

I don't have a good answer for you, but that is an awesome looking setup.
