Just took the mains off this block fresh from machining. I was about to mock up and measure bearing clearance. One of the mains looks like it has a dent or chip. It appears like it may have been done after the align hone, but maybe it’s just a defect?

Is this something that needs to go back for refinishing, or should it be good to go if the clearance is good?

That's a slot for the Tang on the bearing to place it in the right spot. It also acts as a anti-rotation slot.

That little tiny nothing chip??
It's nothing.

As long as it isn't raised material you are fine. Just file it if its high like @eldubb440 stated and you are good to go.
