why does a 727 leak fluid without drive shaft?
 
Because there is an open hole in the trans without the slip yoke.

what year 727 do you have? Slip yoke or ball & trunion?

Probably convertor drain back unless trans is over full.
 
Yeah that's what got me thinking about this Lefty. I wanted to run the engine without a drive shaft. Got the drive shaft in and fluid in the trans but was curious on why trans would leak fluid without a yoke in the end of trans. The yoke doesn't look like it seals anything. I don't know

Rest assured that the yoke is indeed a big part of what seals the back of the trans.... think of the output shaft as "wet", as it is "in the oil" on its spline side, yet the oil seal and the dust seal both ride on the yokes

Yes the rear seal isn't just there for looks.
 
In other words the seal rides on the yoke and only seals when the yoke is in place.
