I am looking to purchase performance 727 and was wondering where i should start? I am running a 440 around 475-500 hp.

maybe try a TCI

The tci super streetfighter TF 727 is rated 600 to 650 horses out of the box and is fairly affordable at about 1100 to 1300,just a suggestion.
 
I run the TCI 727 with a 5500 stall and a 456 gear. The transmission hangs in there just fine. I have had it in the car for 5000 miles. It has put up with many front wheels in the air holeshots and keeps going. That is my 2cents worth.

Phoenix Transmissions in Weatherford Texas will make you whatever you want. They have done trannies for "One Lap" cars, and are the only people who I want to build mine.

Pro Trans in Lancaster CA. If you look at 75% of the stock/ super stock cars have his sticker. He also builds all my transmissions. I can say first hand he is honest and a pleasure to deal with. His transmissions are perfect from the get go and are affordable. 661-940-7400
 
I'm considering the TCI Super myself,but have even considered buying internal components and having a good local guy build mine from what's in the car now. Anyone here use any Mancini Racing trans components or products?

My 66 Charger came with its stock 440 727 trans.
